The full name of MTT is 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (Thiazolyl Blue Tetrazolium Bromide), and the Chinese name is abbreviated: thiazolyl. The molecular formula is C18H16BrN5S and the molecular weight is 414.32.

 

MTT is often used to detect cytotoxicity and cell proliferation. The detection principle is that the succinate dehydrogenase in the mitochondria of living cells can reduce the exogenous MTT to water-insoluble blue-purple crystal formazan (Formazan) and deposit it in the cells. The cells have no such color reaction. Dimethyl sulfoxide (DMSO) can dissolve formazan in cells, and measure its light absorption value with a microplate reader. Within a certain cell number range, the amount of MTT crystal formazan formed is proportional to the number of cells.

 

In the traditional MTT detection method, since the formazan produced is insoluble in water, it is necessary to remove the upper layer of the reaction solution containing the MTT substrate, and then add DMSO to dissolve the formazan. This step is likely to cause repeatability and reproducibility of the experiment. Poor sex. This kit provides a special formazan dissolving agent. There is no need to remove the original MTT substrate-containing reaction solution during detection. Instead, the formazan dissolving agent can be added directly to dissolve formazan, thereby avoiding the removal of the original reaction solution.
