G2020-25ML Ultra Sensitive ECL Chemiluminescence Kit For Electrophoresis gel color development

This product ECL is a high sensitivity chemiluminescence kit, the basic principle is luminol based ECL chemiluminescence. The product can react with horseradish peroxidase (horseradish peroxidase,HRP) coupled with diantibody to produce fluorescence. The samples are further optimized by using X sheet or other appropriate fluorescence imaging equipment (CCD camera, etc.). Higher sensitivity, more stable color, suitable for low abundance target protein detection.

There are two kinds of ECL chemiluminescence kits produced by our company, G2014 ECL chemiluminescence kit and G2020 hypersensitive ECL chemiluminescence kit. The target protein with high abundance, such as internal reference protein, is recommended to be used G2014, detect target protein with content greater than or equal to 2ng. For the target protein with low abundance and difficult to detect, G2020 is recommended, the sensitivity of this product is 5 times higher than that of G2014, and the protein of pg grade can be detected, and the luminescence is more stable.

Technical Specification

Storage condition:

Stored at 4ºC by ice bag(wet ice), avoid direct sunlight, valid for 12 months. If not used for a long time, it can be stored at -20ºC.

 

Usage:

  1. Preparing for hypersensitive ECL Chemiluminescence Kit: mix hypersensitive ECL A solution and hypersensitive ECL B solution, pay attention to 4ºC avoid light preservation. Available for use within 2 days.
  2. Western experiments PVDF membrane was incubated with the second antibody and washed several times. Place two layers of PE gloves or other transparent films on the exposure box, place the protein of the PVDF membrane face up between the two layers of the exposure box, and add a mixed hypersensitive ECL chemiluminescence solution to cover the membrane with 1-2 min.
  3. Use filter paper or absorbent paper to remove hypersensitive ECL chemiluminescence solution and cover the upper film to start pressing the film.
  4. he finished film is developed and fixed with developing solution, fixing solution(recommended G2019,G2023,G2024), etc. Adjust the exposure conditions according to the luminous intensity.

 

Notes:

  1. Hypersensitive ECL A solution and the hypersensitive ECL B solution must replace the suction head in the process of absorption. After the A solution and the B solution pollute each other, the A solution or the B solution will gradually fail. In addition, metal ion pollution will reduce the sensitivity of this reagent. Please pay attention to clean tips. Seal and save after use.
  2. If the background is very deep after exposure, the reason may be that the concentration of the second antibody is too high, or the concentration of the first antibody is too high, or the sealing solution is not suitable, and the other sealing solution is replaced.
  3. If the fluorescence is quenched rapidly, the reason may be that the target band has too strong fluorescence, resulting in rapid depletion of ECL by HRP.
  4. If there is no luminescence signal, the target protein expression may be very weak, which can prolong the pressing time.
  5. Please wear lab cloths and disposable gloves.
