The synthesis and decomposition of proteins in cells are in dynamic balance under stable conditions.However, when proteins are extracted from cells and tissues in vitro, cells contain many endogenous proteases that can degrade proteins, such as phosphatase and serine protease.These ubiquitous endogenous proteases cause the target protein to break down, thereby affecting the experimental results.

To avoid protein degradation, protease inhibitors are usually added to protein extraction. PMSF (Phenylmethanesulfonyl fluoride), with molecular formula C7H7FO2S and molecular weight 174.19, inhibits serine proteases (such as trypsin, chymotrypsin, thrombin) and mercapto proteases (such as papain) in an irreversible way.

Technical Specification

Usage:

  1. The concentration of this product is PMSF 100 mM, and the final concentration of the working liquid is suggested to be 1 mM.
  2. 5 minutes before use, add 10 microliters of this product per milliliter of protein lysate to make the required dosage, and mix for reserve use.

 

Notes:

  1. PMSF is highly toxic and can seriously damage the mucous membranes of the respiratory tract, eyes and skin. It can be fatal if inhaled, swallowed or absorbed through the skin.In case of contact with PMSF through eyes or skin, rinse immediately with plenty of water.
  2. PMSF is unstable in aqueous solution and should be added to the cracking buffer on the spot from the storage solution before use.
  3. Please wear laboratory clothes and gloves during operation.
